Abstract
BACKGROUND
riple-negativebreastcancer(TNBC)isthemostmalignantsubtypeofbreastcancerwithhighlyinvasiveabilityandmetastaticnaturetothelymphnodes.Longnon-codingRNAs(lncRNAs)havebeenwidelyexploredincancertumorigenesisandprogression.However,theirrolesinTNBClymphnodemetastasisremainsrarelystudied.
METHODS
heexpressionoflncRNAhighlyupregulatedinmetastaticTNBC(HUMT)incelllinesandtissueswasdetectedbyquantitativereal-timePCR(qRT-PCR)andinsituhybridization(ISH).RNAimmunoprecipitation(RIP)andRNApulldownwereusedtoverifytheinteractionbetweenlncRNAandprotein.Chromatinimmunoprecipitation(CHIP)anddCas9-gRNA-guidedchromatinimmunoprecipitation(dCas9-CHIP)wereconductedtoidentifythespecificbindingsiteofHUMT-YBX1
